The record
| Class | bone anabolic |
|---|---|
| Also known as | PTH 1-34, Forteo, Bonsity, rhPTH(1-34) |
| Chain length | - |
| Molecular weight | 4,117.8 Da |
| Half-life | ~1 hour (SC); 5 min (IV true elimination) |
| Route | SC (thigh or abdomen) |
| Studied for | Postmenopausal osteoporosis; Male osteoporosis; Glucocorticoid-induced osteoporosis |
| Research use | Bone formation studies; PTH receptor research; Fracture healing |
| What has been shown | Increases BMD 8-10% in spine (1 year); Reduces vertebral fractures 65%; Reduces non-vertebral fractures 53%; Reduces hip fracture risk >50% |
| How it works | Recombinant human parathyroid hormone fragment (amino acids 1-34) that binds to PTH type 1 receptors (GPCRs) on osteoblasts. Intermittent administration preferentially stimulates osteoblastic bone formation over osteoclastic resorption. Activates cAMP and calcium signaling pathways. Daily pulsatile exposure is anabolic; continuous exposure would be catabolic. |
